چکیده

Effects of 6-hydroxy-L-nicotine derived from nicotine catabolism in Arthrobacter nicotinovorans on learning and memory processes were examined in adult male Wistar rats. 6-hydroxy-L-nicotine (0.3 mg/kg, i.p., 7 consecutive days chronic administration) significantly increased spontaneous alternation in Y-maze task and working memory in radial arm-maze task, suggesting effects on short-term memory, without affecting long-term memory, explored by reference memory in radial arm-maze task. Taken together, our results suggest that 6-hydroxy-L-nicotine sustain memory formation and may provide the opportunity to manage neurological abnormalities in neurodegenerative diseases.

Localization of the enantiozymes of 6-hydroxy-nicotine oxidase in Arthrobacter oxidans by electron immunochemistry.

During the course of growth of Arthrobacter oxidans, induction of the enantiozymes 6-hydroxy-D-nicotine oxidase and 6-hydroxy-L-nicotine oxidase occurred in the presence of DL-nicotine. Cryoultramicrotomed sections obtained from cells grown to stationary phase were gold immunolabeled. The results obtained demonstrate that both enzymes are localized in the cytoplasm.

Gene cluster on pAO1 of Arthrobacter nicotinovorans involved in degradation of the plant alkaloid nicotine: cloning, purification, and characterization of 2,6-dihydroxypyridine 3-hydroxylase.

A 27,690-bp gene cluster involved in the degradation of the plant alkaloid nicotine was characterized from the plasmid pAO1 of Arthrobacter nicotinovorans. The genes of the heterotrimeric, molybdopterin cofactor (MoCo)-, flavin adenine dinucleotide (FAD)-, and [Fe-S] cluster-dependent 6-hydroxypseudooxynicotine (ketone) dehydrogenase (KDH) were identified within this cluster. The gene of the la...

The Effect of Ethanolic The effect of ethanolic extract of Saffron (Crocus sativus L.) on improving the spatial memory parameters in the experimental models of Parkinson disease in male rats

Background & Objective: The axial role of the oxidative stress in the pathophysiology of Parkinson disease has been identified. On the other hand, the learning and memory impairment in Parkinson disease has a distinguished outlook. Since Saffron has antioxidative stress effects, the aim of the present study is to investigate the improving effects of Saffron extract on the spatial memory paramet...

Effects of Maternal Separation on Nicotine-Induced Conditioned Place Preference and Later Spatial Learning and Memory Function in Adolescent Male Rats

Background: Disturbances in maternal care have been associated with increased risk for drug abuse later in life. However, there has been little investigation of the effects of maternal separation (MS), a model of early life stress, on nicotine dependence, specifically during adolescence. In the present study, we aimed to investigate the effect of MS on nicotine-conditioned place preference (CPP...

A novel mammalian expression system derived from components coordinating nicotine degradation in arthrobacter nicotinovorans pAO1

We describe the design and detailed characterization of 6-hydroxy-nicotine (6HNic)-adjustable transgene expression (NICE) systems engineered for lentiviral transduction and in vivo modulation of angiogenic responses. Arthrobacter nicotinovorans pAO1 encodes a unique catabolic machinery on its plasmid pAO1, which enables this Gram-positive soil bacterium to use the tobacco alkaloid nicotine as t...
